Bioremediation of oil-polluted soil by Lentinus subnudus, a Nigerian white-rot fungus

Abstract
Inspite of the realization and studies on the use of microorganisms in degrading hydrocarbons there has been very little work on indigenous white- rot fungi in Nigeria, a leading oil – producing country. The ability of Lentinus subnudus to mineralize soil contaminated with various concentrations of crude oil was tested. Organic matter and carbon were higher than the control at all concentrations of crude oil contamination in soils inoculated with L. subnudus for 3 months. Nutrient contents were generally higher after 6 months of incubations except potassium levels which were not significantly different from the control. As for the total petroleum hydrocarbon (TPH) in crude – oil contaminated soils; the highest rate of biodegradation was at 20% concentration after 3 months and 40% after 6 months of incubation.

Aflatoxin B1 production in chillies (Capsicum annuum L.) kept in cold stores

Abstract
An attempt has been made to isolate and enumerate the mycoflora invading chillies kept in cold storage since May, 1999. Chilli pods were collected from the cold stores at monthly intervals for a period of one year between December 2002 to November 2003. The incidence of molds on unsterilized as well as surface sterilized samples was recorded. Species of Aspergillus found to be dominant on stored chillies were screened for antimicrobial activity. The toxins from A. flavus and A. niger exhibited antibacterial and antifungal activity. Incidences of Alternaria alternata, Fusarium spp. and Mucor spp. was low on stored chillies when compared to Aspergillus spp. Chilli seeds aseptically collected from the pods were also tested for mycoflora. Natural occurrence of aflatoxin B1 in chilli pods kept in cold storage was tested. Results from HPLC analysis revealed that the samples were contaminated with aflatoxin B1 to the extent of 5.5 mg Kg-1.

Raw starch degrading amylase production by mixed culture of Aspergillus niger and Saccharomyces cerevisae grown on sorghum pomace

Abstract
Production of raw starch degrading amylase by a mixed culture of Aspergillus niger and Saccharomyces cerevisae grown on sorghum pomace as nutrient source was investigated. Effect of mineral nutrient supplementation of sorghum pomace on raw starch degrading amylase activity was also determined. Sorghum pomace medium significantly (P<0.05) induced higher level of raw and extracellular amylase than soluble starch medium. Mixed culture media recorded higher (P<0.05) level of raw starch degrading amylase than monoculture media. However, mineral nutrient supplementation significantly (P<0.05) suppressed raw starch degrading amylase production. The crude enzyme solution degraded both cereal and tuber or root starches significantly (P<0.05). Sources of crude enzyme significantly (P<0.05) influenced raw starch digesting activity. Optimum pH for the raw starch degrading amylase which varied between 3.0 and 8.0 depended on the source of the crude enzyme.

Bacteria and fungi isolated from housefly (Musca domestica L.) larvae

Abstract
Housefly larvae were cultured on fresh fish and collected for the isolation and identification of microorganisms associated with them. The microbes were cultured from both the gut and body surface of the maggot on nutrient agar (for bacteria) and potato dextrose agar (for fungi) and incubated at about 37°C for 48 h before observations. A variety of microorganisms, which includes the pathogenic Staphylococcus aureus, Pseudomonas aerruginosa, Aspergillus tamarii and Bacullus cereus were found. Also, nonpathogenic microbes werer recovered including Bacillus subtilis and Stroplococcus faecalis.

Production dynamics of extracellular protease from Bacillus species

Abstract
Screening and isolation of proteolytic bacteria were carried out from soil samples of Ikogosi warm spring (SW, Nigeria) . Eighteen isolates were positive on skim milk agar (10%) of which fifteen produced protease in culture broth. Three isolates, identified as Bacillus macerans IKBM-11, B. licheniformis IKBL-17 and B. subtilis IKBS-10, were selected for further study. These Bacillus species could grow up to 65°C within a broad pH range of 5 to 10 with an optimal growth temperature and pH at 60°C and 8.0, respectively. For the three Bacillus species, protease production occurred between 37°C and 65°C and pH 5 to 10. Maximum growth and maximum enzyme production was observed at 48 h when grown in 50 ml medium (pH 8.0) under shaking condition at 60°C. The results showed that Bacillus species under study are good producers of extracellular protease at high temperature. This might be an indication that proteases produced would be thermostable.

Isozyme diversity in Cassia auriculata L.

Abstract
Cassia auriculata is considered to be one of the important dye yielding and medicinal plants in India. In the present study seeds from fourteen different localities were collected all over India and nine enzymes were screened by native polyacrylamide gel electrophoresis (PAGE) technique and thirty-four putative loci were totally detected. Cluster and factor analyses indicated that there are two major distinct groups or clusters, and thus, seeds collected from a few different localities are enough to capture the genetic variation held by this species. Also isozyme analysis is a reliable, efficient and effective marker technology for determining genetic variations in C. auriculata.
