Cadmium and lead levels in some fish species from Azuabie creek in the Bonny Esturary, Nigeria

Abstract

The concentrations of cadmium and lead were determined in seven fish species from the Azuabie creek in the upper Bonny estuary of the Niger Delta, which is associated with industrial and abattoir discharges. Cadmium concentrations ranged from 0.01 to 0.06 mg/kg and show no significant difference between species. The concentrations of lead (ranging from 0.104 to 0.310 mg/kg) were significantly higher in Chrysichthys nigrodigitatus and Gobius niger than other species examined. Also, lead was significantly higher than cadmium. The metal concentrations obtained were within values that may be considered safe for human consumption.

Effect of exogenous progesterone on oestrus response of West African Dwarf (WAD) goats

Abstract

Twenty-four (24) healthy, parous West African dwarf (WAD) does aged 2 – 3 years were used to study the effects of varying doses of progesterone on oestrus synchronization and plasma progesterone levels. The does were randomly assigned to 4 treatment groups consisting of 12.5, 25.0 and 37.5 mg progesterone treatments and 1.0 ml physiological saline as the control. The animals were monitored for signs of oestrus during and after the treatment. Blood samples collection from each treatment groups on days 7 and 14 of progesterone treatment were used for serum progesterone evaluation. The results showed that the number of oestrus goats, percentage oestrus goats and oestrous cycle length were similar between the control and goats on 12.5 mg progesterone injection. The does treated with 12.5 mg progesterone showed longer oestrus duration that differed significantly (P < 0.05) from goats on 25.0 and 37.5 mg progesterone and the control treatment. There was no significant difference (P > 0.05) in intensity of heat between the treatment groups. Although higher serum progesterone levels were observed in goats treated with 37.5 mg progesterone, 12.5 mg was more effective in producing heat at the withdrawal of progesterone treatment. The results of this study indicate that progesterone injection would be effective in synchronizing heat in WAD goats.

Effects of disinfection, packaging and evaporatively cooled storage on sugar content of mango

Abstract 

The present investigation was undertaken to evaluate the effect of post-harvest disinfection, packaging, evaporative cooling storage and their combined effect on the changes in sugar content of mango (Mangifera indica L.). The experiment was laid out in a factorial combination of disinfection, packaging and storage in a randomized complete block design with three replications. The mangoes were periodically analyzed for reducing sugar and total sugar. Non-reducing sugar was computed from the difference between experimental reducing and total sugars. Storage conditions significantly (P ≤ 0.01) affected sugar content in mangoes. Storage at ambient conditions with higher temperature and lower relative humidity as compared to the evaporatively cooled storage resulted in rapid deterioration in sugar content of the mangoes. During the storage period, packaging generally maintained higher (P ≤0.01) levels of reducing sugar, non-reducing sugar and total sugar. Similarly, disinfection treatment significantly (P ≤ 0.01) affected the changes in reducing, non-reducing and total sugars of mangoes during storage. Two-way interactions were significant (P ≤ 0.01) in terms of the changes in sugar content of mangoes. The benefits of the combined effect of post-harvest treatments on mangoes included maintenance of high reducing sugar, non-reducing sugar and total sugar.

High yield and high quality DNA from vegetative and sexual tissues of Mexican white pine (Pinus ayacahuite)

Abstract

Pines are considered to be difficult for DNA extraction. However, from one species to the other there is variation in phenolic profiles and seed size that might affect final DNA yields and quality. Two DNA extraction protocols (CTAB and SDS based) were compared for their ability to produce DNA on leaves, gametophyte and embryo from Pinus ayacahuite, a pine species with a large seed (8 - 18 mm). The DNA obtained from both procedures was quantified and tested by PCR. The CTAB protocol provided higher DNA yields from vegetative tissue and embryo than the SDS method. Embryos (2n) and gametophytes proved to be very good sources of DNA and the DNA isolated was suitable for PCR-RAPD and SSR markers. This paper reports the results and describes the modified CTAB protocol.

Bioremediation of engine-oil polluted soil by Pleurotus tuber-regium Singer, a Nigerian white-rot fungus

Abstract

White-rot fungi have been used in various parts of the world for bioremediation of polluted sites. Pleurotus tuber-regium was noted to have the ability to increase nutrient contents in soils polluted with 1 - 40% engine-oil concentration after six months of incubation. P. tuber-regium increased organic matter, carbon and available potassium 5.19%, 2.99% and 0.97 meq/100 g respectively compared to 4.41%, 2.56% carbon and 0.66 meq/100 g, respectively in the control. However, higher values of 0.32% nitrogen, 11.42 ppm phosphorus and pH 6.94 were obtained in the control compared to 0.16% nitrogen, 9.32 ppm phosphorus and pH 5.93 in soils incubated with the fungus. The fungus brought about an increase in copper content in engine oil polluted soils at 10% concentration followed by a decrease at 20 and 40% concentrations. Bioaccumulation of zinc and nickel was recorded at 20% engine-oil concentrations.
